  1. Product introduction

Recombinant Kex-2 protease (e.c.3.4.21.61) belongs to the subtilisin family. It is a precursor processing enzyme encoded by yeast itself, a Ca2 + – dependent serine protease. It can specifically recognize and cleave the carboxy terminal peptide bond of double basic amino acids such as Lys Arg, Arg Arg and pro arg. Unlike trypsin, this enzyme can not recognize and cleave the carboxy terminal peptide bond of single basic amino acid, namely Arg or Lys. Kex2 protease activity was not inhibited by conventional serine protease inhibitors such as aprotinin, PMSF and TPCK. The action pH is pH9.0, and the stable storage pH is ph5.0-6.0.

This product is obtained by the secretory expression technology of recombinant Pichia pastoris and is a recombinant Kex2 protease with high purity and activity.

  1. Recommended use

L recommended reaction buffer: pH 7.0-9.0, 50mm Tris HCl, 2mm Ca2 + or HEPES, 5mm Ca2 +. If it is not used immediately after dissolution, it is recommended to use 20mm (pH 5.2) NaAc HAC and 2mm Ca2 + buffer to dissolve the lyophilized powder. The final concentration of the enzyme after dissolution is about 1-10mg / ml. it can be stored under – 20 ℃ after sub packaging as required.

L reaction, using pH 7.0-9.0, 50mm Tris HCl, 2mm Ca2 + or HEPES, 5mm Ca2 + as reaction diluent.

Note: the reaction of enzyme is pH9.0, and the stable pH is 5.0-6.0.

 

  1. Transport and storage stability

L transportation stability: ice bag thermal insulation transportation, stable activity.

L storage stability: freeze dried powder is stable within 24 months after sealed storage at – 15 ℃. Dissolve it in storage buffer (20mm NaAc HAC, 2mm Ca2 +, ph5.2), and store it at – 15oC after dissolution. It will be stable for 6 months.
